Abstract FGF/FGFR signaling plays an important role in both embryonic and postnatal skeletal development. Osteoblasts play a leading role in bone formation, and osteoblast differentiation is the precondition of bone formation. FGF/FGFR signaling regulates the expression of various iconic genes in different stages of osteoblast differentiation. The signal can not only effect on osteoblast differentiation by itself, but also interact with other signaling pathways (BMP, Wnt and PTH), mutual coordination regulate osteoblast differentiation. FGFR mutation can cause abnormal osteoblast differentiation, which lead to a variety of bone diseases, such as craniosynostosis, osteoporosis, heterotopic ossification, etc. The following review summarized FGF and FGFR family, the genes and markers of osteoblast differentiation, FGF/FGFR signaling regulating osteoblast differentiation and other aspects.
