Abstract Objective To investigate the role of peptide through biopanning with the target of PGE2 in RA, which can give a new method for treating this disease. Method By using PGE2 as the selective molecule, the T7 select phage was biopanned and positive clones were selected by ELISA and DNA sequencing. After setting up the adjuvant induced arthritis model, we investigated the role of anti-inflammation of synthesized peptide by observing the edema of paws and the quantity of pro-inflammatory cytokines by ELISA. Results After enrichment , among random 23 clones there were 21 positive,among which there had the same sequence and conservative amino. While the peptide can reduce the level of paw edema and the pro-inflammatory cytokines in joint of RA model. Conclusions The method of screening the antagonist of receptor of PGE2 by phage display is applicable, while this peptide can play the anti-inflammation role.
