A Rapid and Direct Detection of Cyclodextrins in the Fermentation Fluid by HPAEC1

Abstract  In order to investigate the formation of various cyclodextrins (CDs) by cyclodextrin glucanotransferase (CGTase), a rapid and direct method was developed.The method was applied to screen the CGTaseproducing bacteria in the soil. Among the 149 strains, which can produce starch hydrolysing enzymes, 11 bacterial strains were detected to produce CDs with HPAEC at the experimental conditions, 6 mainly produced CD6, 5mainly produced CD7, and none could produce CD8 as the main product.In the process of a direct identification of CGTase strains,the method can detect quantitatively the content of various CDs including largering cyclodextrin(CD>8).
