Strategies for high-level expression of recombinant protein in Escherichia coli |

Abstract Cloning and expression of recombinant protein in Escherichia coli has been developed for a long time and it is widely used as a classical expression system now. Many advantages of this system have been confirmed, however, disadvantages also exist. The efficiency of expression is affected by many factors. This paper summarized strategies for high-level expression of recombinant protein in this system. These strategies mainly include selection of appropriate promoter, optimization of signal peptide, increasing the stability of mRNA and the efficiency of translation, preventing the use of low usage codons, reducing inclusion body formation and degradation of the target protein, using molecular chaperones and fusion protein, development of high-cell-density fermentation, and etc.
